Can one assign new mates in an assembly using the Isolate command?

When an assembly has zero mates assign, is one able to use the 'Isolate' command to assign new mate connections without moving the parts of the isolated portion of the assembly?
Can anyone provide one example of this? Google search retrieved no examples on this idea.

Yes, one thing to keep in mind is that it's helpful to select the 2 items you want to mate and "isolate". You also don't want to enter isolate before inserting a new part (or pasting a part) as any part added whit isolate active is excluded (which seems odd...).
Note that you should be able to enter "isolate" while the mate dialogue is open so you can also select your first mate connector and then enter isolate.
Don't forget that you also have the "transparent" option, which basically is the reverse of isolate (i.e. if you were to invert you selection and use isolate it would result in the same as hitting isolate).0
